人类干细胞miRNAPCR芯片用于研究干细胞感应、维护、分化和识别相关的84 miRNA的表达。所有的干细胞都能够自我更新和分化成多种细胞类型。多能干细胞能够分化成3胚层的任何类型细胞,而3胚层表达一组对多能性至关重要的miRNA,包括可以提高重编程使分化细胞回到干细胞状态。然而,最近的研究表明, 与胚胎干细胞(ESC)相比,诱导多能干细胞(iPSC)和始发态(或外胚层)干细胞(epiSC)miRNA表达有差异。在分化过程,多能性标记物的表达水平下降而其他分化关键miRNA的表达增加。附加的miRNA在特定阶段或在特定血统变得更加普遍,导致miRNA对三种不同胚层祖细胞独特的表达模式。这个芯片确定miRNA的相对表达对多能性和分化干细胞重要性。芯片还额外包括代表胚胎干细胞(ESC),诱导多能干细胞(iPSC),外胚层干细胞(epiSC),胚状体的身体细胞,胚层祖细胞的标记。每张芯片含一个对照组使得分析数据时可以用相对定量ΔΔCT方法评估逆转录效率,基因组DNA污染,PCR效率。利用这张芯片,通过实时定量PCR,可以简易且可靠地检测与干细胞最相关的miRNA的表达。
